摘要
目的观察尾吊模拟失重对大鼠颈总动脉和股动脉自噬活性的影响。方法雄性SD大鼠,随机分为尾吊组和对照组,通过Western Blot、PCR及免疫组化检测尾吊模拟失重后大鼠颈总动脉及股动脉微管相关蛋白轻链3(LC3)和Bcl-2相关蛋白(Beclin-1)的表达变化。结果尾吊28 d后,大鼠股动脉LC3及Beclin-1的mRNA和蛋白的表达均显著高于对照组,免疫组化发现LC3在内皮层及平滑肌层均有表达升高;而颈总动脉的LC3及Beclin-1的mRNA和蛋白的表达无明显变化。结论尾吊模拟失重可以诱导大鼠股动脉自噬活性增强。
Objective To investigate the changes of autophagy activity in the carotid arteries and femoral arteries of rats with tail suspension. Methods Male Sprague-Dawley rats were randomly divided into control and simulated weightlessness groups. Tail suspension for 28 d was used to simulate microgravity in this study. The expression of Microtubules-associated protein 1A light chain 3( LC3) and Bcl-2 interacting protein( Beclin-1)were detected by Western blot and RT-PCR in all groups. Moreover,immunohistochemistry assay were performed to evaluate the source of autophagy changes in vascular. Results Compared with the control group,the expression of mRNA and protein of Beclin 1 and LC3 had no significant change in the carotid artery of the simulated microgravity group,while Beclin 1 and LC3 expressions were significantly enhanced in the femoral arteries. Moreover,immunohistochemistry result showed that the expression of LC3 was increased in the endothelial layer and smooth muscle layer of the femoral artery. Conclusion The autophagy activity in the femoral arteries of rats under simulated weightlessness,tail suspension,for 28 d is stimulated,but not in the carotid artery.
