摘要
目的探讨小分子干扰RNA(si RNA)下调星形细胞上调基因1(AEG-1)表达对胃癌细胞SGC-7901侵袭和迁移能力的影响及可能机制。方法将AEG-1 si RNA及阴性对照分别转染SGC-7901细胞,q RT-PCR及Western blot-ting检测转染后AEG-1m RNA及蛋白的表达,细胞划痕实验检测SGC-7901细胞迁移能力,Transwell实验检测SGC-7901细胞侵袭能力,Western blotting检测转染后侵袭相关蛋白MMP2、MMP9及VEGF的表达。结果转染AEG-1 si R-NA后,SGC-7901细胞AEG-1m RNA及蛋白表达明显下调(P<0.05),迁移和侵袭能力显著降低(P<0.05),MMP2、MMP9及VEGF蛋白表达水平明显下调(P<0.05)。结论 AEG-1 si RNA可抑制SGC-7901细胞迁移和侵袭,其机制可能与抑制侵袭相关蛋白的表达相关。
Objective To explore the effect of down-regulation of AEG-1 siRNA on the invasion and migration abilitiesof gastric cancer cell line SGC-7901. Methods Gastric cancer cell line SGC-7901 was transfected by AEG-1 siRNA andnegative control respectively. q RT-PCR and Western blotting were performed to measure the expression of AEG-1 m RNA andprotein. Wound healing assay and Transwell assay were applied to investigate cell migration and invasion respectively. Theexpression of MMP2、MMP9 and VEGF were measured by western blotting. Results After transfection of AEG- 1 siRNA,AEG-1 m RNA was down-regulated in SGC-7901 cells(P〈0.05),and cell invasion and migration abilities were decreased(P〈0.05),as well as the expression of MMP2、MMP9 and VEGF(P〈0.05). Conclusion AEG- 1 siRNA may suppress cellinvasion and migrationin SGC-7901 cells,the mechanism may be related to inhibit invasion related protein expression.
