不同核酸提取试剂在肠道病毒核酸检测中的比较

Comparison of different nucleic acid extraction reagents in the detection of enterovirus

目的比较国产试剂与进口试剂提取手足口病病例标本肠道病毒核酸的重复性、灵敏度及对核酸的回收率。方法用国产磁珠法试剂盒与进口离心柱法试剂盒同时提取手足口病病例标本、用于检测和作为阳性质控品的细胞培养物,通过荧光PCR方法检测病毒核酸,比较2种核酸提取试剂所提取的病毒核酸的重复性、灵敏度及对核酸的回收率。结果国产磁珠法与进口离心柱法重复性、灵敏度无明显差异;国产磁珠法回收率为80.02%,进口离心柱法回收率为75.63%。结论国产试剂与进口试剂性能无明显差异;国产磁珠法较离心柱法方便快捷,国产试剂可替代进口试剂用于手足口病病原学检测。

objective To compare the repeatability, sensitivity and recovery rate of extracting nuclear acids betweenhomemade and imported extracting reagents. Methods The homemade and imported extracting reagents were used to extractnucleic acid from specimens of HFMD patients and cell cultures. The repeatability, sensitivity, and recovery rate of the tworeagents were compared by using Q-PCR. Results The repeatability and sensitivity of the two reagents were not significantlydifferent, the recovery rates of homemade reagent and imported reagents were 80.02% and 75.63%, respectively. Conclusion The repeatability and sensitivity of the two reagents are not significantly different in nucleic acid isolation. However, thehomemade reagent is more convenient and quick, it is alternative to imported extracting reagent in detection of pathogenof HFMD.
