湖南汝城县热水温泉产嗜热琼胶酶和淀粉酶菌的筛选及鉴定 被引量:1

摘要 目的对汝城热水镇温泉中产嗜热琼胶酶和淀粉酶菌株进行筛选及鉴定,为其在医学方面的潜在价值提供实验依据。方法采用琼胶酶培养基、碘-淀粉培养基的透明圈法,结合H/C比值筛选高产嗜热琼胶酶和淀粉酶的菌株,从形态观察、理化性质和16S r DNA基因序列分析对其进行鉴定。结果筛选出3株降解嗜热琼胶酶、嗜热淀粉酶能力较高的菌株R8、R11、R13;其中通过H/C比值计算出R13降解琼胶的能力最强为34;降解琼胶能力强弱的菌株为:R13>R11>R8。R11菌株水解淀粉的能力最强为18,且三株菌株水解淀粉能力的强弱为R11>R8>R13。3株菌经16S r DNA初步鉴定均为土芽孢杆菌属。结论本研究所获得的具有产较高活性的嗜热琼胶酶和淀粉酶菌株,提示其在医学方面有深入的研究空间。 Objective To provide the experiment basis for its hidden value in Medicine through screening and identificating bacterium producing agar enzyme and starch enzyme in HuMan RuCheng hot spring. Methods Transparent circle method with agar enzyme and iodine-starch medium was adoptted, along with H/C to screen high-product bacterium which produced agar enzyme and starch enzyme. And then identify it by its appearance, physico-chemical property and 16S rDNA gene sequencing. Results Three bacteria were screened ( R8,R11 ,R13 ) with higher ability on degradating thermophilic agar enzyme and starch enzyme. By method of H/C, it was found that R13 degrades agar's ability was strongest up to thirty-four, degradating agar's ability from strong to weak was R13〉Rll〉RS. Rll amylatic ability was the strongest up to eighteen, and amylatic ability from strong to weak was R11 〉RS〉R13. Three bacteria were initially identified as Geobacillus through 16S rDNA. Conclusion Highly active thermophilic agar enzyme and starch enzyme bacteria produced in the study show significant value in a further study on Medicine.
