PDGF-BB对大鼠阴茎海绵体平滑肌细胞增殖和细胞周期的影响

The effects of PDGF-BB on cell proliferation and cell cycle of corpus cavemosum smooth muscle cells in SD rat

摘要 目的探讨血小板源性生长因子BB(PDGF-BB)对大鼠阴茎海绵体平滑肌细胞(CCSMC)增殖和周期的影响。方法使用改良组织块贴壁法原代培养大鼠阴茎海绵体平滑肌细胞,将细胞分为空白对照组和不同浓度PDGF-BB处理组,分别采用MTS法和流式细胞术检NPDGF-BB对CCSMC增殖和周期的影响。结果MTS法提示PDGF-BB促进CCSMC增殖,且在一定时间范围和浓度范围内分别呈时间依赖和浓度依赖;流式细胞术结果显示PDGF-BB作用后G0/G1期细胞由(90.79±3.46)%减少至(77.57±5.72)%;而S期的细胞由(7.85±2.68)%升高至(16.47±4.42)%;G2/M期由(2.46±1.25)%升高至(5.96±2.18)%,两组间差异具统计学意义(P〈0.05)。结论PDGF-BB可促进大鼠阴茎海绵体平滑肌细胞增殖。 Objective To evaluate the effect of the platelet-derived growth factor-BB (PDGF-BB) on cell proliferation and cell cycle of corpus cavemosum smooth muscle cells (CCSMCs) in SD rat. Methods CCSMCs were primarily cultured and subjected to immunoeytochemical assay. The cells were divided into different concentrations of PDGF-BB groups and a nromal control group. The MTS method and flow cytometry were used respectively to detect cell proliferation and cycle. Results MTS analysis suggested PDGF-BB increased CCSMC proliferation, and exhibited time dependent and concentration dependent mode; Flow cytometry results showed that the proportion of cells in G0/G1 phase decreased from (90.79±3.46)% to (77.57±5.72)%; And the proportion of cells in S phase was increased from (7.85±2.68)% to (16.47±4.42)%; G2/M phase cells from (2.46±1.25)% to (5.96±2.18)%, there were significant differences between the two groups (P〈0.05). Conclusion PDGF-BB may promote rat corpus cavernosum smooth muscle cell proliferation.
