The G protein-coupled estrogen receptor(GPER), a novel estrogen receptor that differs from classical estrogen receptors(ERs), is discovered in the late 1990 s. Since then, GPER is gaining attention and widely research. More and more evidence has indicated that GPER is involved in the occurrence and development of cancer and cardiovascular disease, and may represent a new target to combat these disease. While recent reports regarding the location and function of GPER on the growth of cancer cells are controversial. In this review, the intrinsic appearance of GPER, including structure, subcellular localization, ligands and signal transduction pathway is simply introduced, the role of GPER in various cells proliferation and its relationship with diseases are mainly discussed in order to further understand the GPER and its roles in signal transduction, cell proliferation and disease prevention and treatment.
