摘要
蛋白质多肽激素是一大类内源信号分子,通过激活靶细胞膜上特定受体发挥重要调控功能。配基-受体结合测定是研究蛋白质多肽激素与受体相互作用的重要实验方法,但传统方法要使用放射性同位素(如碘-125),限制了该方法的广泛使用。我们在近期工作中利用超灵敏的Nano Luc荧光素酶建立了"生物发光配基-受体结合测定新方法"用于研究蛋白质多肽激素与受体的相互作用。该方法具有灵敏度高、重复性好、安全无害、操作简便等的优点,有望广泛用于蛋白质多肽激素与受体相互作用研究。
Protein or peptide hormones are a large group of signaling molecules and play important biological functions by binding specific cell membrane receptors. Ligand-receptor binding assay is an important technique to study the interactions of protein or peptide hormones with their receptors. However, the conventional assays rely on radioisotopes, such as radioactive iodine-125, and thus their applications are restricted. In recent studies, we have developed novel bioluminescent ligand-receptor binding assays for some protein or peptide hormones based on the ultrasensitive bioluminescence of a small Nano Luc reporter. The novel bioluminescent ligand-receptor binding assay is ultrasensitive, safe and convenient, thus it would be applied to more protein or peptide hormones in future studies.
