摘要
目的研究自动化平板接种仪对副溶血性弧菌国家标准检测方法的改进效果,以期提高副溶血性弧菌国家标准检测方法的阳性检出率和检测效率。方法从副溶血性弧菌阳性检出率较高的天然海产品样品中选取70份样品,使用自动化平板接种仪和国家标准法,对样品进行对比检测。结果使用仪器法检出22份副溶血性弧菌阳性样品,阳性检出率为31.4%。国家标准法检出20份副溶血性弧菌阳性样品,阳性检出率为28.6%。两者差异无统计学意义(P>0.05),但仪器法阳性数多2份,而且在实际操作中检测效率有所提高。仪器法相较国家标准法平板上单个分离菌株生长率高,仅有5份样品需要再次转种纯化,比国家标准法减少了31个工作日。结论自动化平板接种仪可用于副溶血性弧菌的检测,能够提高检测效率,缩短检测时间,减少操作误差。
Objective To make a research the improvement of national detection standard in the detection of Vibrio parahaemolyticus with automated petri dish inoculated instruments,in order to improve its positive detection rate and detection efficiency.Methods 70 samples were selected from natural seafoods with higher positive rate of Vibrio parahaemolyticus. Automated petri dish inoculated instruments and the national standard method were used for samples test and comparison. Results Using the automated petri dish inoculated instruments,22 Vibrio parahaemolyticus were detected,with the positive rate of 31. 4%. Using the national standard,20 Vibrio parahaemolyticus were detected,with the positive rate of 28. 6%. Although there was no statistical significance on the difference between the two methods( P〈0. 05),but there were more than two cases of positive samples with automated petri dish inoculated instruments,and it improved the detection efficiency in actual operation. Compared to the national standard method,a single strain isolated growth rates were higher on the plate with automated petri dish inoculated instruments,only 5 samples required two purifications. It reduced 31 working days,compared to the national standard method.Conclusion This automated petri dish inoculated instruments can be used for the detection of Vibrio parahaemolyticus,it can improve the detection efficiency,shorten detection time,and reduce operator error.
