原发性干燥综合征患者血液中miR-146a-5p、miR-4484和miR-4687-5p的表达 被引量:5

Expression of miR- 146- a- 5p,miR- 4484 and miR- 4687- 5p in the blood of patients with PSS

摘要 目的通过检测3种原发性干燥综合征(PSS)相关的miRNA(miR-4687-3p、miR-4484、miR-146a-5p)在PSS患者和健康人群中的差异表达水平,探讨其在PSS诊断和临床检验中的作用和意义。方法采用荧光定量PCR(q PCR)法检测28例患者和30例健康志愿者血清中3种miRNA的差异表达水平,利用诊断价值评价使用受试者工作特征曲线(ROC)分析miRNA的敏感性和特异性。结果经过q PCR验证,PSS治疗前组与对照组比较miR-146a-5p、miR-4484和miR-4687-5p表达均上调(P<0.05),治疗前组和治疗后组比较,miR-146a-5p和miR-4484表达均下调(P<0.05),而miR-4687-5p表达依然上调(P=0.993>0.05)。3种miRNA的灵敏性和特异性分别为miR-4484>miR-4687-5p>miR-146a-5p。结论 miR-4687-3p、miR-4484、miR-146a-5p表达异常可能参与了PSS的发生和发展,miR-4484可能成为PSS的早期辅助诊断指标。 Objective By measuring the differential expression levels of three miRNA( miR- 4687- 3p,miR- 4484,miR-146- a- 5p) associated with primary sjogren's syndrome( PSS) in PSS patients and healthy people,so as to explore its role and significance in the diagnosis and clinical application of PSS. Methods Fluorescence quantitative PCR( q PCR) method was used to detect the differential expression levels of three kinds of miRNA in 28 cases and 30 healthy controls cases,using the diagnosis value evaluation and the receiver- operating characteristic curve( ROC) to analyze the sensitivity and specificity of miRNA. Results After q PCR verification,miR- 146a- 5p,miR- 4484 and miR- 4687- 5p expression were raised in PSS pre- treatment group,compared with control group( P〈0. 05). Comparing the group before treatment and after treatment,miR- 146a- 5p,miR- 4484 expression were reduced( P〈0. 05),while miR- 4687- 5p expression was still raised( P =0. 993 0. 05). Sensitivity and specificity of the three kinds of micrornas were miR- 4484 miR- 4687- 5p miR-146a- 5p. Conclusion The abnormal expression of miR- 4687- 3p,miR- 4484,miR- 146- a- 5p may be involved in the occurrence and development of PSS,miR- 4484 will be the early auxiliary diagnosis index for PSS.
