不同剂型七味白术散对腹泻小鼠肠道菌群失调和肠粘膜紧密连接蛋白的影响 被引量:22

Effects of Different Formulated Qiwei Baizhu Powder on Intestinal Flora and Tight Junction Protein in Diarrhea Mice

摘要 目的研究七味白术散,七味白术滴剂和七味白术提取物对抗生素联合番泻叶致腹泻小鼠肠道菌群失调和肠粘膜连接蛋白的影响。方法小鼠经头孢曲松钠和盐酸林可霉素加冰番泻叶灌胃造成菌群失调模型。将小鼠分正常组(N G)、模型组(M G)[造模成功后即为自然恢复组(ZH G)]、七味白术散煎剂组(QJ G)、七味白术滴剂组(QD G)、七味白术提取物组(QT G)。治疗后,进行粪便含微生物种类鉴别及数量测定和肠粘膜紧密连接蛋白(Claiidin-1,Occludin和ZO-l)的荧光密度分析。结果 QD G和QT G中肠道中肠球菌、肠杆菌、乳杆菌和双歧杆菌四种菌群菌量均有一定幅度增加,与自然恢复相比,差异有统计学意义(P<0.05或P<0.01);QJ G和ZH G菌量虽有增加,但不如QD G和QT G恢复迅速。药物治疗后,小肠绒毛尖端紧密连接蛋白Claudin-l,Occludin和ZO-1的荧光密度明显增高,绒毛尖端和紧密连接部破坏减轻,说明各药物能促进受损肠粘膜的修复。结论七味白术散能够调节肠道菌群失调和促进肠粘膜损伤修复。 Objective To investigate effects of Qiwei Baizhu Powder,Qiwei Baizhu Drops and Qiwei Baizhu extract on intestinal flora and tight junction protein in diarrhea mice with antibiotics jointed senna. Methods With ceftriaxone,lincomycin hydrochloride and cold-senna gavaging mice to make intestinal flora,and model animals were were divided into 5 groups: model group( M G)( model success is natural recovery group( ZH G)),normal group( N G),Qiwei Baizhu decoction group( QJ G),Qiwei Baizhu Drops group( QD G),Qiwei Baizhu extract group( QT G). Analysis of intestinal microbial species identification and quantity in feces and fluorescence intensity of mucosal tight junction protein( Claudin-1,Occludin and ZO-l) were taken at 5days,7 days,9 days,11 days,13 days after treatment. Results Compared with ZH G,the quantity of enterococci,Escherichia coli,Lactobacillus and Bifidobacterium bacteria increased significantly( P〈0. 05 or P〈0. 01); QJ G or ZH G also increased the amount of bacteria,but not as good as QD G and QT G rapidly. After treatment,fluorescence intensity of Claudin-l,Occludin and Z0-1 in the tight junction protein of villus tips significantly increasing,villus tip and the tight connecting portion damage attenuating,it showed that each drug can promote the repair of damaged intestinal mucosa. Conclusion Qiwei Baizhu Powder can regulate intestinal flora and promote intestinal damage repair.
