摘要
目的考察梅花鹿血浆中IGF-1贮存稳定性。方法应用建立的ELISA检测方法对在不同保存方法下鹿血IGF-1稳定性进行研究。结果鹿血浆室温(20~25℃)贮存1~3天,IGF-1损失较少;在冷藏(4℃)的条件下贮存1~7天,IGF-1损失也较少;在冷冻(-20℃)条件下贮存,半年内IGF-1损失率变化不大;反复冻融对血浆中IGF-1的稳定性影响较大,冻融1次就损失近30%。结论为提高鹿血浆中IGF-1的利用率,建议及时对其进行处理或冷藏的条件下贮存(7天内),否则冷冻贮存,避免反复冻融。
Objective To explore the stability of IGF-1 in deer blood at different preservation conditions. Methods The established ELISA method was applied for the detection of IGF-1 in deer blood at different preservation conditions. Results The loss rate of IGF-1 was relatively low,if deer blood was stored at room temperature for 1 to 3 days,the IGF-1 losses decreased slowly when saved in refrigerated 4℃ for 7 days. Kept in-20℃ for six months,the loss rate of IGF-1 was little. But repeated freezing and thawing had a great effect on IGF-1 stability. The loss rate of IGF-1 would be 30%,even if repeated freezing and thawing was only once. Conclusion In order to increase utilization rate of IGF-1 in deer blood,it was suggested that deer blood should be processed on time,keep in cold storage less than 7 days,or keep in freezing storage less than 6 months. The repeated freezing and thawing should be avoided.
