脾阳虚证导致的大鼠心功能变化及其机制 被引量:2

Changes of cardiac functions in rats with spleen-yang deficiency pattern and its underlying mechanism

摘要 目的探讨脾阳虚对大鼠心功能的影响及其机制。方法将30只SD大鼠随机分为2组,分别为空白对照组及脾阳虚模型组。采用饮食不节加力竭游泳再加灌服蕃泻叶法制备脾阳虚证大鼠模型。采用小动物超声测定2组大鼠的心功能变化;Western blot法及免疫组化法测定心肌中脑钠肽(BNP)的蛋白表达情况;ELISA法测定血清及心肌中BNP、环磷酸腺苷(c AMP)含量;RT-PCR测定成碱性成纤维细胞生长因子(b FGF)、蛋白激酶A(PKA)mRNA的表达。结果与空白组比较,脾阳虚组大鼠心输出量及射血分数均降低(P<0.05),血清及心肌中BNP、c AMP含量均升高(P<0.05),BNP蛋白表达及b FGF、PKA mRNA表达均有所增加(P<0.05)。结论脾阳虚可引起大鼠心功能下降,与体内BNP、c AMP含量及PKA、b FGF表达增加有关。 Objective To explore the changes of cardiac functions in rats with spleen-yang deficiency pattern and its underlying mechanism. Methods 30 Sprage Dawley( SD) rats were randomly divided into two groups: the blank control group and spleen-yang deficiency model group. After successful modeling with improper diet,exhaustive swimming and intragastric gavage of senna leaf( Fanxieye),changes of cardiac functions in the two groups were determined by using ultrasonic imaging with a high-resolution in vivo imaging system. Protein expression of brain natriuretic peptide( BNP) in the myocardium was assessed by using Western blotting and immunohistochemistry. Contents of BNP and cyclic adenosine monophosphate( c AMP) in the serum and myocardium were measured with Enzyme-linked Immuno Sorbent Assay( ELISA). The mRNA expression of Basic Fibroblast Growth Factor( b FGF) and protein kinase A( PKA) were detected by using Reverse Transcription-polymerase Chain Reaction( RT-PCR).Results Compared with the blank control group,Stroke Volume( SV) and Ejection Fraction( EF) decreased( P〈0. 05) while the contents of c AMP and BNP in serum and myocardium increased( P 0.05). BNP protein expression and mRNA expression of b FGF and PKA increased( P〈0. 05) in the model group. Conclusion Cardiac function seems to decline in spleen-yang deficiency rats. Contents of BNP and c AMP,and expression of PKA and b FGF all appears to have increased in the model group.
