羌活不同地理种群cpDNA trnT-trnL多态性分析 被引量:4

Polymorphism analysis of endangered Notopterygium incisum and endemic species based on cpDNA trnT-trnL sequences

摘要 目的对245份不同产地羌活Notopterygium incisum样品的遗传多样性和遗传结构进行研究。方法采用叶绿体cpDNA trnT-trnL直接测序的方法和邻接法(NJ)对245份羌活样品进行遗传多样性和遗传结构分析。结果物种水平上单倍型多样性(Hd)指数为0.873,核苷酸多样性(Pi)指数为0.004 07;种群水平上Hd介于0~0.900,Pi指数介于0~0.054 4,与同属的宽叶羌活Notopterygium franchetii相比,遗传多样性水平居中;分子方差分析表明,羌活大部分遗传变异(77.55%)发生在居群间,居群间的基因流(Nm=0.145)较低,群体间变异是羌活的主要变异来源;NJ树表明,受试的31个野生种群可分为2大类群。结论羌活的遗传多样性处于中等水平,居群间遗传分化较大。 Objective To analyze the genetic diversity and genetic structure of 245 plant samples in Notopterygium incisum from different regions. Methods Genetic diversity and genetic structure of 245 samples of N. incisum were studied using chloroplast cpDNA trnT-trnL direct sequencing method and Neighbor-joining(NJ) method. Results Haplotype diversity(Hd) and nucleotide diversity(Pi) at the species level were 0.873 and 0.004 07, respectively. At the population level, Hd ranged with 0.000—0.900, and Pi ranged with 0.000—0.054 4. It indicated that N. incisum had a moderate level of genetic diversity than N. franchetii plants of the genus Notopterygium Boiss. Molecular variance analysis(AMOVA) indicated that high genetic differentiation(77.55%) existed among populations, and gene flow was low(Nm = 0.145) among populations. Based on the NJ tree, the 31 wild populations tested were clustered into two groups. Conclusion N. incisum has the midium cp DNA diversity and higher population genetic differentiation.
