复合组织工程人工骨修复兔骨缺损的实验研究

摘要 目的探讨体外复合人工骨对兔骨缺损的修复及干细胞增殖、凋亡情况。方法将实验兔分为A(自体骨植入组)、B(复合人工骨植入组)、C(空白组)3组,体外培养兔骨骨髓基质干细胞复合人工骨材料,按组并分时间段植入兔桡骨骨缺损模型中,分时段取材进行免疫组化、反射学、拉伸应力实验、大体观察、光镜检查等获取体外复合人工骨对兔骨缺损的修复情况,及人工骨粘附干细胞增殖、凋亡情况。结果术后4周时B组在成骨速度、成骨质量及抗拉伸强度较B、C组快、强。术后8周B组在成骨速度、成骨质量及抗拉伸强度均仍较A组强,但差距在减少,C组仍无明显骨痂。术后12周A组在成骨速度、成骨质量及抗拉伸强度和B组无明显区别,C组术后12周骨折边缘模糊,少量骨痂形成。实验A组BMP-2平均灰度值与对照B组比较有统计学意义(P<0.05),与对照组C组比较有统计学意义(P<0.05),对照B组与对照C组比较有统计学意义(P<0.05)。结论复合骨组织材料有利于兔成骨细胞的增殖、粘附和分化,能促进成骨细胞在体外成骨的能力,在体内可降解且具有较好的骨传导能力。 Objective Investigate the compound artificial bone of rabbit bone defect repair and stem cell proliferation and apoptosis.Methods Experimental rabbit can be divided into A(autologous bone implant group)and B(composite artificial bone implant group),C(blank group)three groups,in vitro culture of rabbit bone marrow stromal stem cells composite artificial bone material,according to the group and A timebox inserted in the rabbit model of radial bone defect,period of time based on immunohistochemical,reflexology,tensile stress test,gross observation,light microscope examination to obtain composite artificial bone in vitro in rabbit bone defect repair case,and artificial bone adhesion stem cell proliferation and apoptosis.Results 4 weeks after surgery in group B in osteogenesis,osteogenesis quality and tensile strength at a B and C group fast and strong.After eight weeks of group B in osteogenesis speed,the quality of osteogenesis,and tensile strength is still relatively strong group A,but the gap in decline,still no obvious callus of group C.12 weeks after surgery in group A osteogenesis speed,quality of osteogenesis and tensile strength and no significant difference between group B and group C after 12 weeks of fracture edge blur,A small amount of callus formation.The experimental group A BMP-2 average grey value compared with control group B(P〈0.05),compared with the control group C(P〈0.05),control group B compared with control group C(P〈0.05).Conclusion Composite bone tissue of rabbit osteoblasts proliferation,adhesion and differentiation,can promote the ability of osteoblast in vitro osteogenesis,biodegradable in the body and has good bone conduction ability.
