TLR4、TBK/IRF3信号通路介导肾小管上皮细胞与单核细胞相互作用 被引量:4

摘要 目的:探讨单核细胞与肾小管上皮细胞相互作用的分子机制。方法:体外建立单核细胞(U937)与肾小管上皮细胞(HK-2)共培养系统,观察HK-2细胞对U937细胞Toll样受体4(toll-like receptors 4,TLR4)及TANK结合蛋白激酶(TBK)/干扰素调节因子3(IRF3)信号通路的影响;观察阻断TBK/IRF3信号通路对单核细胞分化的影响。结果:HK-2细胞促进U937细胞TLR4及TBK/IRF3信号通路蛋白及mRNA的表达(P<0.05);阻断TBK/IRF3信号通路抑制HK-2细胞诱导U937细胞发生M1型转化(P<0.05)。结论:TLR4分子介导HK-2细胞与U937细胞相互作用,激活TBK/IRF3信号通路,导致U937细胞发生M1型转化,进一步诱导炎症因子产生,促进肾脏纤维化。
