糖尿病肾病合并抗Thy-1肾炎模型构建及p27^(kip1)的变化 被引量:1

摘要 目的:构建糖尿病肾病合并抗Thy-1肾炎模型,观察模型的发病及病理变化,探讨p27kip1在模型中的变化及意义。方法:将大鼠随机分为4组:正常组(NC组,n=10),糖尿病肾病组(DN组,n=10),抗Thy-1肾炎模型组(Thy组,n=10),糖尿病肾病合并抗Thy-1肾炎模型组(DN+Thy组,n=10)。分别留取2周、4周、6周、8周大鼠24 h尿测尿蛋白定量,8周后留取肾脏组织分别用于提取RNA、蛋白和肾脏病理检查。结果:(1)24 h尿蛋白定量:2周以后各模型组与NC组相比尿蛋白增多(P<0.05),DN+Thy组与DN组、Thy组相比尿蛋白增多(P<0.05);(2)各模型组p27kip1mRNA表达量的变化,DN组的明显增加;Thy组降低;DN+Thy组的表达量略增加,但与正常组比差异无统计学意义。结论:(1)成功诱导出糖尿病肾病合并抗Thy-1肾炎的动物模型;(2)肾皮质细胞周期负调控蛋白p27kip1mRNA表达量的变化为DN合并抗Thy-1肾炎与DN、抗Thy-1肾炎的鉴别提供依据。
