摘要
目的:观察不同浓度的葡萄糖对MG-63细胞增殖、分化及成骨能力的影响。方法:实验分为正常糖浓度组(5.5mmol/L)、高糖组(15.5mmol/L)、超高糖组(25.5mmol/L)三组糖浓度。甲基噻唑基四唑(MTT)法测定1d,3d,5d的细胞增殖数;碱性磷酸酶(ALP)活性检测1d,3d,5d,7d的细胞分化情况;茜素红染色观察14d,21d细胞分泌钙结节的情况。结果:随糖浓度增大,MG-63细胞数量、ALP活性、钙结节数目的表达均显著降低,差异有统计学意义(P<0.05)。结论:高糖能显著抑制MG-63成骨细胞的增殖、分化。
Objective: To observe the influences of different concentration of glucose on proliferation、differentiation and osteogenic capability of MG-63 osteoblasts. Methods: Experiment are divided into three glucose concentrations (5.5mmol/L, 15.5mmol/L, 25.5mmol/L). MTT assay to detect the number of the osteoblasts at 1d、3d、5d, the activity of alkaline phosphatase (ALP) to detect the differentiation of osteoblast at 1d、3d、5d、7d, the alizarin red dye to observe the number of calcium nodus at 14d, 21d. Results: Along with the increased glucose concentrations, there was significant decrease of cell numbers, ALP activity, calcium nodus. Conclusions: The high glucose concentrations are significantly inhibit the proliferation, ALP activity and mineralization of MG-63 osteoblasts.
