高尿酸血症大鼠的血清脂肪酸代谢特征及祛浊通痹方干预研究 被引量:4

摘要 [目的]通过气相色谱-质谱(gas chromatography-mass spectrometry, GC-MS)联用技术探究高尿酸血症大鼠血清脂肪酸代谢变化及祛浊通痹方对其影响。[方法]以高脂饲料饲喂法建立高尿酸血症大鼠模型,分别采用别嘌醇和祛浊通痹方干预3周,收集末次给药24h后的大鼠全血,利用GC-MS对所有大鼠血清中31种脂肪酸含量进行检测,以单因素方差分析比较不同组间脂肪酸含量的差异。[结果]与正常(N)组比较,模型(M)组大鼠血尿酸水平显著升高,伴随单不饱和脂肪酸总量显著下降,棕榈酸、棕榈油酸、油酸含量显著降低,均有统计学差异(P<0.05)。与M组比较,别嘌醇和祛浊通痹方干预后,别嘌醇(T1)组和祛浊通痹方(T2)组大鼠血尿酸水平显著降低(P<0.05),上述脂肪酸含量则升高(P<0.05)。与N组比较,T1组亚麻酸、神经酸含量显著降低(P<0.05),而T2组亚麻酸、神经酸含量无统计学差异(P>0.05)。[结论]高尿酸血症大鼠存在脂肪酸代谢紊乱,尤其是单不饱和脂肪酸下调明显;别嘌醇和祛浊通痹方均可改善高尿酸血症大鼠的脂肪酸代谢,而且祛浊通痹方对亚麻酸和神经酸等脂肪酸含量无明显影响。 [Objective]To explore the changes of serum fatty acid metabolism in hyperuricemia rats and the effect of Quzhuo Tongbi decoction by gas chromatography-mass spectrometry(GC-MS).[Methods]Hyperuricemia model was established by high fat diet feeding method.The hyperuricemia rats were intervened with allopurinol(T1 group)and Quzhuo Tongbi decoction(T2 group)for 3 weeks,respectively.The whole blood was collected 24 hours after the last administration.The contents of 31 fatty acids were measured by GC-MS,and the differences among different groups were compared by oneway ANOVA.[Results]Compared with normal(N)group,the level of serum uric acid in model(M)group increased significantly,and the content of total monounsaturated fatty acids decreased(P<0.05).Especially,the contents of palmitic acid,palmitoleic acid and oleic acid decreased significantly(P<0.05).Compared with M group,the levels of serum uric acid in T1 group and T2 group were lower,and the contents of above fatty acids were higher(P<0.05).Compared with N group,the content of linolenic acid and nervonic acid in T1 group decreased significantly(P<0.05),while there was no significant difference in the content of linolenic acid and nervonic acid in T2 group(P>0.05).[Conclusion]There was disturbance of fatty acid metabolism in hyperuricemia rats,especially the decrease of monounsaturated fatty acids.Both allopurinol and Quzhuo Tongbi decoction can improve the fatty acid metabolism of hyperuricemia rats,and Quzhuo Tongbi decoction has no effect on content of linolenic acid and nervonic acid.
