摘要
目的:建立HPLC法测定重楼中4种甾体皂苷的含量。方法:采用色谱柱为Thermo BDS Hypersil C_(18)(250 mm×4.6 mm,5μm),流动相为乙腈-水梯度洗脱,流速0.8 ml·min^(-1),检测波长为205 nm,柱温为25℃。结果:4种重楼皂苷的标准曲线线性良好(r>0.999 5,n=5),重楼皂苷Ⅰ、Ⅱ、Ⅵ、Ⅶ的平均回收率为100.2%,99.93%,97.61%,99.33%;RSD分别为1.29%,1.21%,2.13%,1.84%(n=6);不同批次重楼中4种重楼皂苷的含量有一定差异。结论:该方法准确、简单、快捷,可用于重楼中4种皂苷的定量分析。
Objective: To establish a simultaneous determination method for four steroidal saponins in Paris ployphyUa by HPLC. Methods: The column was Thermo BDS Hypersil C18 (250 mm ×4.6 mm,5 μm), the mobile phase was acetonitrile-water with gradient elution at a flow rate of 0.8 ml · min-1. The detection wavelength was 205 nm. The column temperature was 25℃. Results: The calibration curves of four steroidal saponins showed good linear relationship (r 〉 0.999 5 ). The recovery of steroidal saponin Ⅰ,Ⅱ, Ⅵ and Ⅶ was 100.2%, 99.93%, 97.61% and 99.33%, respectively. The RSD was 1.29%, 1.21%, 2.13% and 1.84% (n =6), respectively. The results showed that the contents of the four steroidal saponins in Paris ployphylla were various among different batches. Conclusion: The method is accurate and convenient, and suitable for the quantitative analysis on four steroidal saponins in Paris ployphylla.
