脂肪干细胞的生物学特性及其在骨组织工程中的研究进展

摘要 组织和器官的修复与重建,是生物学和临床医学面临的重大难题之一,而干细胞治疗为再生医学提供了新思路。脂肪干细胞是一类存在于脂肪组织中,具有与骨髓间充质干细胞类似的自我更新和多向分化潜能的成体干细胞,并逐渐成为组织工程的种子细胞。本文对脂肪干细胞的生物学特性进行了综述,阐述了脂肪干细胞的表型、多向分化潜能、培养扩增方法及多种疾病的临床前实验成果,分析了脂肪干细胞作为干细胞来源的可行性,并且总结了脂肪干细胞在骨组织工程中的研究现状和发展前景。 Repair and reconstruction of tissue and organ is one of the major problems in biology and clinical medicine.Stem cell therapy provides a new idea for regenerative medicine.Adipose-derived stem cells(ADSCs) are in adipose tissue,have self-renewal and differentiation potentials of adult stem cells,similar to bone marrow stem cells,and gradually become the seed cells for tissue engineering.This paper reviews the biological characteristics of ADSCs,elaborates phenotype,differentiation potential,culture and amplification methods of ADSCs,and preclinical experiments of certain diseases.The feasibility of ADSCs as a source of stem cells is analyzed,and the research status and development prospects of ADSCs in bone tissue engineering are summarized.
