摘要
目的建立山茱萸醇提物的高效液相(HPLC)指纹图谱,对所获得的指纹图谱信息进行聚类分析。方法山茱萸药材10批,用50%乙醇料液比为1∶8,回流提取3次每次3 h,浓缩,用甲醇稀释。应用Accurasil C18高效液相色谱柱(250 mm×4.6mm,5μm),二极管阵列检测器(DAD),以0.15%磷酸溶液和乙腈为流动相,梯度洗脱,流速1 m L·min-1,柱温30℃,检测波长217 nm。结果山茱萸样品中共检测到12个共有色谱峰,相似度在0.748~0.919之间。用聚类分析法进行分析,将样品归类。用主成分分析法进行分析,可见样品6相对于其它9个样品得分最高,样本性质或特征的相似程度最好。结论建立的指纹图谱具有稳定、重复性好的特点,可作为山萸肉骨质疏松效应成分物质基础研究的有效手段。
Objective To establish the fingerprint of Cornus officinalis by HPLC. Methods The 10 batches of drug underwent circumfluence extraction 3 times using 50% ethanol: material = 1 ∶ 8 as the extracting solution,and 3 hour per time. Then the extracts were,concentrated,diluted with methanol. Subsequently,the sample solution was separated on an Accurasil C18 column( 4. 6 mm × 250 mm,5 μm) and detected by DAD. Mobile phase: aqueous phosphoric acid( 0. 15%) and acetonitrile. Analysis was performed at 30℃ with a flow rate of 1 m L·min- 1and UV detection at 217 nm.
