摘要
目的通过对各组绒毛给予不同浓度的睾酮,检测用药低浓度组与用药高浓度组绒毛HCG和VEGF的分泌情况,探讨睾酮对体外培养人早孕胚胎绒毛分泌hCG和VEGF的影响,进一步探讨临床上高雄激素血症对妊娠的影响。方法取人早孕绒毛,行体外组织块培养法,培养绒毛。分为对照组、加低浓度睾酮组(睾酮浓度0.5ng/ml,0.7ng/ml,1.0ng/ml),加高浓度睾酮组(睾酮浓度1.5ng/ml,2.0ng/ml,2.5ng/ml),ELISA法检测绒毛培养液内hCG,VEGF浓度。比较不同浓度睾酮培养液的绒毛所产生hCG和VEGF与对照组的差值。结果 HCG组检测结果:每个绒毛的组间比较,低浓度组与高浓度组hCG浓度差异有统计学意义(P<0.05);VEGF组检测结果:每个绒毛的组间比较,低浓度组与高浓度组hCG浓度差异无统计学意义(P>0.05)。结论睾酮可通过对绒毛分泌hCG能力的影响进而影响到绒毛功能,进一步导致各种妊娠疾病的产生。
Objective To discuss pregnant women clinical manifestations of hyperandrogenism,pregnancy disease occurrence rate is higher than normal pregnant women;to investigate in vitro cell culture increased with the concentration of testosterone in the fluid,in cultured human early chorionic hCG and VEGF secretion is affected.Methods First,the clinical investigation pregnant women with hyperandrogenism clinical manifestations of the same age healthy pregnant women during pregnancy complication rate of occurrence;the second step,to take people villi and cultured in vitro tissue culture.Divided into control group,plus low levels of testosterone group(0.5ng/ml,0.7ng/ml,1.0ng/ml),heightening levels of testosterone group(1.5ng/ml,2.0ng/ml,2.5ng/ml),within the detection medium hair hCG and VEGF concentration by ELISA.Results HCG group test results:generation difference plus low levels of testosterone and high levels of testosterone group hCG group was statistically significant(P〈0.05);VEGF group test results:group differences in VEGF concentration and a high concentration of low-dose group was not statistically significant(P〉0.05).Conclusion Description testosterone may in turn affect the villi function through its effects on the secretion of hCG fluff ability to further lead to various diseases of pregnancy.
