摘要
目的观察臭氧水对骨科感染性创面β-内酰胺酶大肠埃希菌的体外灭菌效果。方法制取2.5μg/ml、5μg/ml和10μg/ml臭氧水对β-内酰胺酶大肠埃希菌体外定量灭菌一定时间,经中和作用后再培养24 h,计算灭菌率;10μg/ml臭氧水对其体外灭菌作用相应时间,经中和、离心、染色后,观察菌体超显微结构变化情况。结果 2.5μg/ml、5μg/ml和10μg/ml臭氧水0.5 min时对产β-内酰胺酶大肠埃希菌的平均灭菌率分别为17.7%、27.8%和43.7%;1 min时分别为42.9%、72.4%和80.3%;2 min时分别为53.3%、93.5%和96.5%;4 min时分别为59.7%、98.8%和99.8%;8 min分别为76.2%、100%和100%;16 min分别为97.4%、100%和100%。未经过臭氧水作用的产β-内酰胺酶大肠埃希菌菌体椭圆、扁平、边缘整齐光滑、有鞭毛;作用2 min时菌体染色加深、鞭毛脱落断裂;作用4 min时细胞壁塌陷破坏;作用8 min、16 min时菌体皱缩近似圆形、染色出现不均、失去固有扁平的形态。结论臭氧水对产β-内酰胺酶大肠埃希菌有较好的杀灭效果,且这种作用与臭氧水浓度和灭菌时间呈相关性。10μg/ml臭氧水是一种高效快速杀菌剂,可用于骨科创面感染产β-内酰胺酶大肠埃希菌的局部治疗。
Objective To observe the role of the disinfecting effect of ozonated water solution on Escherichia coli- ESBL- producer on wound in orthopedics. Methods The concentrations of 2. 5 μg / ml,5 μg / ml and 10 μg / ml ozonated water were used to disinfect the Escherichia coli- ESBL- producer suspension in vitro quantitative sterilization time,then cultivated again after neutralization and calculated the sterilization rate. The 10 μg / ml of ozonated water disinfected on Escherichia coli- ESBL- producer suspension in vitro at corresponding time was used to observe its ultramicroscopic structure change after neutralizing,centrifuging and dyeing. Results The sterilization rate of ozonated water of 2. 5 μg /ml,5 μg / ml and 10 μg / ml at 0. 5 min on Escherichia coli- ESBL- producer were 17. 7%,27. 8% and 43. 7%; 42. 9%,72. 4% and 80. 3%at 1 min; 53. 3%,93. 5% and 96. 5% at 2 min; 59. 7%,98. 8% and 99. 8% at 4 min; 76. 2%,100% and 100% at 8 min; 97. 4%,100%and 100% at 16 min. After disinfection treatment with the 10 μg / ml ozonated water,its ultrastructural become blurred,surface presented folds,deeper,shrinkage,flagella off fracture. Conclusion The ozonated water was highly effective inactivation on Escherichia coli- ESBL- producer.The role correlated with the ozonated water concentration and sterilization time. The 10 μg / ml ozonated water is an effective and rapid fungicide,which can be used for bone wound infection local treatment of Escherichia coli- ESBL- producer.
