基于肾藏志理论的肾虚质大鼠生长发育与NMDA受体表达实验研究

Experimential Study about Growth and Development and NMDA Receptor Expression of Rats with Kidney-dificiency Constitution on Theory of Kidney Storing Aspiration

摘要 目的:观察肾虚质大鼠生长发育情况和海马N-甲基-D-天冬氨酸(NMDA)受体表达,探讨脑功能与肾藏志理论相关性。方法:采用"猫吓鼠"经典造模法造成先天不足加后天失养复合型肾虚质模型仔鼠,分为模型组、左归丸组、右归丸组,空白组产自正常孕鼠。选用补肾中药进行调理,观察仔鼠各项生长发育指标并检测NMDA受体表达。结果:(1)生长发育情况:与空白组相比,模型组体重下降,毛发稀疏、尿量增多、精神不振,各参数积分值有显著性差异(P<0.05);药物干预后情况明显改善,与模型组比较有统计学意义(P<0.05)。(2)NMDA受体表达:模型组亚基NR2A、NR2B灰度值较空白组低(P<0.05);用药后表达增强,灰度值变化有统计学意义(P<0.05)。结论:肾虚质大鼠在生长发育方面表现为体重下降、毛发不荣、尿量增多,精神不振,且生长发育低下状态与海马NMDA受体表达呈正相关;补肾药物可增强NMDA表达水平同时改善肾虚质大鼠低下的生长发育状况。 Objective:Observe the growth and development and NMDA receptor expression of rats with kidney-deficiency constitution,then disscuss the correlation between brain function and theory of kidney storing aspiration. Methods:By the way of "cat scare rat",built composite offspring rat models with deficiency and acquired dystrophy,then divided them into model group,Zuogui Pill group and YouguiPill group,and blank group offspring came from normal pregnant rats. After the treatment with kidneysupplementing drugs,tested growth and development indexes and NMDA receptor expression. Results:(1)Growth and development situation:Compared model group with blank group,weight declined,hair thinned,urine volume increased and spirit depressed(P〈0.05);after treatment,all of these indexes were better than model group(P〈0.05).(2)NMDA receptor expression:the gray value of NR2 A and NR2 B of model group were lower than blank group(P〈0.05);by the treatment with kidney-supplementing drugs,they increased than model group(P〈0.05). Conclusion:In terms of growth and development,weight of rats with kidney-deficiency constitution loss,hair thin,urine volume increase and spirit depress. All of these declining situations relates with NMDA low expression. Drugs for kidney supplement can increase NMDA expression and improve their low growth and development.
