回补维生素E对维生素E缺乏不孕大鼠妊娠结局的影响 被引量:6

The effect of vitamin E supplement on the pregnancy outcomes of vitamin E-deficiency-associated infertile female rat

摘要 目的研究在植入过程回补维生素E对维生素E缺乏不孕(VED)大鼠妊娠结局的影响。方法从出生后11d开始喂养维生素E缺乏的食物,喂养6个月后建立VED大鼠模型。在模型大鼠交配后5.5d、6.5d、7.5d分别灌胃500mg/kg的维生素E回补,以非VED大鼠为对照组,在回补0h、4h、8h、12h、16h、20h和24h观察维生素E的浓度变化,并于交配后19.5d统计大鼠子宫的着床点个数、活胎个数和胎鼠体重变化。结果 VED大鼠在回补4h后血浆中维生素E的浓度为13.54±3.81μg/ml,达到正常水平;分别在交配后5.5d、6.5d、7.5d回补维生素E的VED大鼠妊娠率分别为80%、60%、40%(对照组均为100%),着床点个数分别为9.3±1.1、8.5±0.5和8.4±1.4(对照组为12.7±0.5、13.0±2.2和12.7±1.2),活胎个数分别为5.0±2.0、4.0±2.3和2.0±2.4(对照组为12.3±0.5、12.7±2.4和11.3±1.7),均显著低于对照组(P<0.05);交配后5.5d回补组胎鼠体重与对照组相比无统计学差异(P>0.05),交配后6.5d和7.5d回补组胎鼠体重显著低于对照组(分别为1.74±0.55g和1.63±0.68g vs.2.16±0.18g和2.19±0.21g)(P<0.01)。结论在植入过程回补维生素E可部分挽救由于维生素E缺乏所导致的不孕;随着回补时间的延后,回补维生素E对妊娠结局挽救的效果明显下降。 Objectives. To study the effect of vitamin E (VE) supplement during the implantation process on pregnancy outcome of VE deficiency-associated infertile female rat. Methods: Female rats were on VE-deficient diet to establish the VE deficiency-associated infertility model (VED group). The model rats (VED group) were given VE (500 mg/kg) by intragastric administration on 5.5,6.5 and 7.5 days post-coitum (dpc) respectively,with the non-VED rat (on normal diet) supplemented with vehicle as control. Plasma VE levels were determined at 0 h,4 h,8 h,12 h,16 h, 20 h and 24 h after supplement. The pregnancy outcomes were determined on 19.5 dpc,with number of implantation sites, live fetus and weight of fetus examined. Results: Plasma VE levels in VED group reached the normal levels at 4 h after VE supplement. The pregnancy rates in VED group with VE supplement at 3 diferent time-points (subgroups 5.5,6.5 and 7.5 dpc) were 80%,60% and 400% respectively (100% in control),the number of implantation sites were 9.3±1.1,8.5±0.5 and 8.4±1.4 respectively (in control,12.7±0.5,13.0±2.2 and 12.7±1.2),and the number of live fetus were 5.0+2.0,4.0±2.3 and 2.0±2.4 respectively (in control, 12.3±0.5,12.7± 2.4 and 11.3±1.7). There was a significant difference in all observed parameters between VED group and control group at all subgroups (P 〈 0. 05). Compared with that in control group, fetus weight was significantly reduced in VED group with VE supplement at 6.5,7.5 dpc (1. 74±0. 55 g,1. 63±0.68 g vs. 2. 16±0.18 g,2. 19±0.21 g) (P〈0.01) ,while no significant difference was found between VED subgroup of 5.5 dpc supplement and normal control(P〈0.05). Conclusion: VE supplement during the implantation process can partially rescue the infertility caused by VE deficiency in rat. The efficacy of rescue was getting lower with the time delayed.
