早产儿不同浓度氧暴露后外周血单个核细胞内p47phox蛋白与活性氧产生的相关性研究

摘要 目的观察早产儿不同浓度氧暴露后,外周血单个核细胞(PBMCs)内p47phox蛋白变化与活性氧(ROS)产生的相关性。方法 32周以下早产儿不同浓度氧疗48 h后,分离PBMCs,检测PBMCs内p47phox的定位情况、p47phox蛋白表达量及ROS的生成量。结果 p47phox随着吸氧浓度升高向细胞膜移位增加;p47phox的蛋白表达量及ROS生成量也随着吸氧浓度的升高而增加。结论早产儿氧暴露后,p47phox可能通过更多的向胞膜移位,增加其表达量来引起ROS产生的增加。 [Objective]To observe the correlation of p7 phox and ROS in PBMCs after different concentration oxygen exposure in preterm infants. [Methods]After oxygen therapy for 48 h in extremely premature infants. We isolated PBMCs from peripheral blood. Then detected the location and expression of p47 phox and ROS within PBMCs.[Results]The higher oxygen concentrated, the more PBMCs have p47 phox translocate to membrane, the expression of p7 phox and ROS production raised as well. [Conclusion]After oxygen therapy, p47 phox may increase the translocation, up-regulate expression to cause reactive oxygen species increasing.
