摘要
目的探讨根尖乳头干细胞(stem cells from the apical papilla,SCAP)在无支架三维培养条件下的生物学行为。方法用显微镜和HR染色观察SCAP球的大体形态,组织学结构;用免疫组化法检测SCAP球中骨涎蛋白(bone sialoprotein,BSP)和血管性血友病因子(von Willebrand factor,vWF)的表达。结果细胞球的大小对球体内部细胞的生长有影响,SCAP球具有向成骨和向内皮分化的潜能。结论无支架三维培养是一种可行的SCAP培养方法。
Objective To study the biology of stem cells from the apical papill (SCAP) in scaffold-free 3D culture. Methods Microscope and HE staining were performed to examine the morphology and structure of the SCAP spheroids. Immunohistochemistry for bone sialoprotein (BSP) and yon Willebrand factor (vWF) was carried out to confirm the contributions of the transplanted SCAP. Results The diameter of the spheroids could affect the vitality of the internal cells; the SCAP spheroids had the potential to differentiation into osteogenesis and endothelial cells. Conclusion Scaffold-free 3D cultured can keep the stem cell biological characteristics of SCAP.
