低强度脉冲超声对阿糖胞苷所致新西兰兔白细胞、血小板减少的影响 被引量:4

摘要 目的:探讨低强度脉冲超声(low intensity pulsed ultrasound,LIPUS)对阿糖胞苷所致新西兰兔白细胞、血小板减少的影响。方法:将28只新西兰兔随机分为2组:正常组(n=8)和模型组(n=20),模型组耳缘静脉注射阿糖胞苷(50 mg/kg),连续3 d。再将模型组随机分为2组:LIPUS辐照组(n=10)和对照组(n=10),LIPUS辐照组(辐照参数0.2 W/cm2;0.6 MHz)辐照7 d。于给药前、给药后、辐照后3、5、7、11、14、17、21 d测定各组新西兰兔白细胞、血小板计数;白细胞分类计数;血小板活化代谢功能。结果:(1)模型组新西兰兔外周血白细胞、血小板与注射阿糖胞苷后降低(t=10.770,P=0.000;t=6.751,P=0.000)。(2)辐照后LIPUS组白细胞计数、中性粒细胞于辐照后5 d开始上升,与正常组比较无统计学意义(P=0.777、P=0.993),与对照组相比上升明显(P=0.017、P=0.028);LIPUS组淋巴细胞计数于辐照后3 d开始上升,但较正常组与对照组差异均不显著(P=0.304、P=0.519);LIPUS组单核细胞计数于辐照后上下波动,与对照组差异均无统计学意义(P=0.926、P=0.855、P=0.858、P=0.255、P18=0.760、P=0.746、P=0.644);(3)辐照后LIPUS组血小板计数上下波动,趋势与对照组一致(P=0.198、P=0.610、P=0.021、P=0.291、P18=0.465、P=0.777、P=0.008)。(4)辐照后LIPUS组血清兔血栓素B2(thromboxane,TXB2)含量较对照组差异不明显(P=0.725、P=0.096)。(5)辐照后LIPUS组骨髓有核细胞数量增加,且辐照处皮肤、肌肉未见异常。结论:低强度脉冲超声可提高阿糖胞苷化疗后白细胞计数,可能是因为其能升高化疗后中性粒细胞数量所致。低强度脉冲超声可能对血小板数量及血清TXB2无影响。 Objectlve :To observe the influence of low intensity pulsed ultrasound (LIPUS) on chemotherapy-induced leucopenia and chemotherapy-induced thrombocytopenia by Ara-C in rabbit. Methods:Totally 28 rabbits were divided randomly into normal group (n=8) and model group(n=20). Ara-C(50 mg/kg) was injected into rabbits in model group from the ear vein for 3 days. Then rabbits in model group were randomized into LIPUS group(n=10,0.2 W/cm2,0.6 MHz) and control group(n=10);the cell number of white blood cell(WBC) ,platelet(PET), NE, LY, MO and thromboxane(TXB2) were counted. Results : ( 1)The cell number of WBC and PLT of rabbits in model group decreased after Ara-C injection(t=lO.770,P=O.OOO;t=6.751 ,P=0.000). (2)The cell number of WBC and NE of rabbits in LIPUS group increased compared with that of control group from 5th day after treatment(P=O.017;P=O.028);the cellnumber of LY of rabbits in LIPUS group increased from 3th day after treatment, but there was no significant difference compared that of with normal and control group(P=O.304;P=0.519). The cell number of MO of rabbits in LIPUS group were fluctuated, but there was no significant difference compared with that of control group (P=0.926; P=0.855 ; P=0.858 ; P=0.255 ; P=0.760; P=0.746;P=0.644). (3)The cell number of PLT of rabbits inLIPUS group were fluctuated, and there was no significant difference compared with that of control group ( P=0. 198 ; P=0.610; P=0.021 ; p=0.291 ;P=0.465 ;P=0.777 ;P=0.008). (4)There was no significant difference in the serum TXB2 levels of rabbits between control group and LIPUS group (P=0.725;P=0.096). (5)The nucleated cells number of bone marrow were increased. The skin and muscle had not been seen abnormal after treatment. Conclusion : LIPUS could increase the cell number of WBC, which may be related with its increase of cell number of NE. LIPUS had no effect on the number of platelets and the serum TXB2.
