摘要
为鉴定河北省谷子病穗中分离获得的一个滑刃线虫群体,对其相关形态特征及测量值进行比较,并对其核糖体28S rRNA-D2/D3区与ITS区核酸序列进行比对和系统进化分析。结果表明:从河北谷子中分离的滑刃线虫群体在形态上与以前报道的贝西滑刃线虫群体非常相似;与Gen Bank中贝西滑刃线虫序列相比,28S rRNA-D2/D3区序列相似度为100%,ITS序列相似度为98%~99%;系统进化分析表明,河北谷子中贝西滑刃线虫群体与Gen Bank中贝西滑刃线虫以100%支持率聚类于同一分支。因此,河北谷子病穗中分离获取的滑刃线虫群体为贝西滑刃线虫。
To identify a nematode isolate from infected ears of millet in Hebei province,the morphological characteristics and measurements of the nematode isolate were acquired and compared,its 28 S rRNA-D2 / D3 and ITS regions of ribosomal RNA were amplified,sequenced and aligned,and the phylogenetic trees were constructed by Neighbor-joining method using MEGA6. 0 software. The results showed: this population was very similar to other Aphelenchoides besseyi populations reported before; compared with the sequences from Gen Bank,the 28 S rRNA-D2 /D3 regions of the nematode isolate was 100% similar to Aphelenchoides besseyi and the similarity of ITS was 98%-99%. The NJ trees showed that the nematode isolate was clustered into one branch with Aphelenchoides besseyi with100% support rate. All the information revealed the nematode isolate from infected ears of millet in Hebei province should be Aphelenchoides besseyi.
