摘要
将低分子量壳聚糖(LMCS)及透明质酸(HA)引入到海藻酸钙(SA-Ca^(2+))凝胶体系中,采用物理交联法制备复合水凝胶珠,并对其溶胀性能及释药性能进行了研究。以苦参碱为模型药物,通过高效液相色谱法(HPLC)考察了该复合凝胶珠的载药及释药行为。结果显示,复合凝胶珠的溶胀率可达46,溶胀时间超过24h。苦参碱质量浓度为3~60μg/mL时与峰面积积分值线性关系良好(r=0.999 6),平均加样回收率为101.34%,日间相对标准偏差(RSD)小于2%,48h后苦参碱的累积释药率为57%,说明该复合凝胶珠对药物具有缓释作用。
Low molecular weight chitosan(LMCS)and hyaluronic acid(HA)were introduced into Ca^(2+)/sodium alginate gel system to prepare the composite gel beads by physical crosslinking method and the swelling properties and drug release properties were studied.Using matrine as a model drug,the drug release behaviors of the composite gels beads were investigated by high-performance liquid chromatography(HPLC)method.Results showed that the swelling rate of composite gel beads was 46 and swelling time was lasted for more than 24 h.The linear relationship between matrine mass concentration(3~60μg/mL)and peak area value was good(r = 0.999 6).The average sample recovery was 101.34% and relative standard deviation(RSD)was less than 2% in one day.After 48 h,the cumulative drug release rate of matrine was about 57%,which indicated that the composite gel beads had sustained-release effect on oral drugs.
