基于微卫星标记的养殖施氏鲟亲鱼群体遗传多样性 被引量:3

摘要 为了解施氏鲟养殖场后备亲本的遗传背景,抑制种质资源退化,防止近交衰退,采用30对鲟属微卫星分子标记,分析施氏鲟后备亲鱼的遗传多样性,筛选出多态性微卫星标记引物。将筛选的引物进行荧光标记,对32尾后备亲鱼进行基因组DNA扩增,采用毛细管电泳进行基因分型,统计分析等位基因数、多态信息含量(PIC)等遗传多样性指标。结果表明:筛选出多态性微卫星标记13对基因,13个位点共扩增等位基因166个,平均等位基因数为12.77,平均有效等位基因数为8.03,等位基因频率为0.006 7~0.4314,平均期望杂合度为0.860 7,平均多态信息含量为0.844 9。初步表明,该亲鱼群体具有较高的遗传多样性。 In order to acquire detailed genetic background of reserved A.schrenckii in sturgeon hatchery,and avoid the degradation of germplasm resources and prevent inbreeding despression,thirty microsatellite loci derived from Acipenser were applied to analyze the genetic diversity for the reserved A.schrenckiiin the hatchery.STR typing of 32 reserved A.schrenckii was analyzed by capillary electrophoresis with those 13 markers which produced by fluorescence label,and the diversity index like the number of alleles(Na)and polymorphism information content(PIC)were calculated and analyzed by statistic method.Results:Thirteen microsatellite markers were amplified polymorphisms for the genomic DNA of reserved A.schrenckii.A total of 166 alleles were detected for 13 loci,the average number of alleles was 12.77,the average effective number of alleles was 8.03,allelic frequency ranged in 0.006 7~0.431 4,the average excepted heterozygosity(He)was 0.8607,and the average polymorphism information content(PIC)was 0.8449.All of these indices indicated that genetic diversity of reserved broodstocks was rich.
