伪狂犬病病毒HNX株增殖特性及其致病性的研究 被引量:2

Proliferation characteristics and pathogenicity of pseudorabies virus HNX strain

摘要 将伪狂犬病病毒(PRV)HNX株和经典Ea株分别接种PK-15和Vero细胞,观察细胞病变情况,用TCID50法检测HNX株的病毒效价并绘制一步生长曲线。将15头50日龄的PRV阴性仔猪随机平均分为3组,其中2组分别滴鼻接种1mL 107 TCID50的HNX株和Ea株病毒,第3组滴鼻接种1mL DMEM培养液作为空白对照。结果显示,HNX株和Ea株在这2种细胞中产生的CPE差异明显。病毒一步生长曲线显示,HNX株早期增殖速度明显比Ea株快。动物试验结果显示,接种HNX株的试验猪临床症状典型,体温和排毒量明显高于Ea株组,HNX株的致死率为60%(3/5),Ea株的致死率为0。HNX株造成的病理损伤比Ea株严重。结果表明,HNX株能快速适应宿主细胞,相关增殖特性发生明显改变,并且对仔猪的致病性明显增强。 This experiment was conducted to study the proliferation characteristics and pathogenicity of pseudorabies virus HNX strain.Cytopathic effect(CPE)was measured on PK-15 and Vero cells infected with HNX and Ea strain,respectively.Viral titer was determined by TCID50 and one-step growth curves were conducted.Fifteen 50-day-old PRV-free piglets were randomly divided into 3groups.The three groups were intranasally inoculated with 107 TCID50 HNX,Ea virus and 1 mL DMEM,respectively.In result,difference in CPE was obviously observed in the HNX group compared with Ea group.One-step growth curves showed that the HNX proliferated significantly faster than Ea in early stage.Typical clinical symptoms were observed in piglets of the HNX group.Body temperature and virus shedding were significantly higher in the HNX group than the Ea group.The mortality of the HNX group was 60%(3/5),however all piglets survived in the Ea group.Pathological damage caused by HNX was severer than Ea.In conclusion,HNX could rapidly adapt to the host cells and the proliferation characteristics changed obviously.More importantly,the pathogenicity of HNX to piglets was significantly enhanced.
