摘要
为阐明绒山羊毛囊超微结构在透射电镜下的识别依据,在绒山羊一个生长周期内,每个月月初活体采集皮肤样品,经固定后制作超薄切片,利用透射电镜拍照和分析。结果显示,绒山羊毛囊超微结构随生长周期的变化而出现不同特征的变化。在毛囊发育早期髓质细胞内可见透明毛质颗粒,细胞表面伸出伪足以桥粒相连,后期毛干形成后髓质细胞发生程序性死亡;毛囊发育中期内根鞘由亨勒层、赫胥黎层和内根鞘小皮由内而外呈同心圆排列,亨勒层为一层角化的扁细胞,在毛囊所有的结构中角质化出现的最早,毛囊发育早晚期内根鞘三层结构不完整。外根鞘毛囊上部和中部为多层细胞,呈正方形,在毛囊的底部为单层,呈扁平型;结缔组织鞘中富含胶原纤维、弹性纤维,感觉神经末梢和血管,在毛囊生长周期内结构变化不大。毛囊发育晚期结缔组织鞘和外根鞘分离。明确不同生长时期毛囊超微结构的特征,能够准确地对生长中的毛囊各层进行识别和定位。
The objective of this study is to provide the important distinguish gist when the microstructure of hair follicle was analyzed by transmission electron microscopy.The ultrathin slice of cashmere goats were made each month in a whole year and were further photographed and analyzed under transmission electron microscopy after staining.In result,each layer histomorphology of the hair follicle of Hexi cashmere goat showed a special characteristic change within a year.In the early development of hair follicles,a large number of transparent hair cells was found in the medulla cells,and programmed cell death was occurred after the formation of the hair stem in the late stage.The inner root sheath was concentrically arranged by Huxley,Henle's layer and cuticle of inner root sheath in the development metaphase of hair follicle,but in the early and late stage the ultrastructure of inner root sheath was not intergal.Henle's layer keratinized earlier than any other layer of inner root sheath.In the upper and middle position,the cells of outer root sheath was multi-layers and square arrangement.There was a single layer cells at the bottom of the hair follicle which is flat arrangement.There were some collagen fiber,elastic fiber,sensory nerve endings and blood vessels in the connective tissue sheath,and no special changes was found in a cycle development of hair follicle.Connective root sheath was separated from outer root sheath in the late stage of hair follicle.Recognition of the ultrastructures of the hair follicle of cashmere goats under the transmission electron microscope could identify and localize exactly to the layers of hair follicle according to the morphological characteristics of developing hair follicle.
