摘要
目的探讨不同粒径的纳米硫化铅对雄性大鼠生殖能力的影响。方法将24只清洁级SD雄性大鼠随机分为4组,分为对照(生理盐水)组和15、35、75 nm纳米硫化铅染毒组,每组6只。采用灌胃方式进行染毒,染毒用量为10 ml/kg,每天1次,连续染毒8周。测定大鼠的精子存活率,并对睾丸进行病理学观察。结果与对照组相比,不同粒径纳米硫化铅染毒组大鼠的精子存活率均较低,差异有统计学意义(P<0.05);且随着纳米硫化铅粒径的增大,大鼠的精子存活率呈上升趋势。病理学观察结果显示,75 nm纳米硫化铅染毒组生精小管变化不明显,各级精母细胞略显减少;35 nm纳米硫化铅染毒组生精小管内各级精母细胞数量减少;15 nm纳米硫化铅染毒组生精小管萎缩呈空网状,管腔内生精细胞脱落坏死。结论纳米硫化铅可损伤大鼠睾丸组织,影响精子生成,具有雄性生殖毒性,且粒径越小毒性越大。
Objective To study the effects of nano-lead sulfide(nano-Pb S) with different particle sizes on reproductive system of male rats. Methods A total of 24 clean grade SD male rats were randomly divided into four groups, namely the control group(saline) and three treated groups(5,35,75 nm granule diameter of nano-lead sulfide),six rats in each group. The rats of treated groups were exposed to nano-Pb S through gavage at the doses of 10 mg/kg body weight, once a day, for eight weeks.The sperm survival rate were detected,the pathological changes of testicle were also investigated. Results Compared with control group, the sperm survival rates of rats in treated groups were all decreased(P〈0.05), and the sperm survival rate increased with the increase of granule diameter of nano-Pb S. Pathological examination showed that the seminiferous tubules changed not obviously in the 75 nm group,spermatogenic cells had a little reduction; It was significantly reduced in the 35 nm group; Most of the spermatogenic cells were necrotic and seminiferous tubules had hollow cross in 15 nm group. Conclusion Nano-Pb S may damage testicular tissue and affect sperm production of male rats. The smaller the particle size of nano-Pb S,the greater the toxicity to the body is.
