摘要
以精品肝素为原料,研究磁化水结合过氧化氢制备低分子肝素(LMWH,2~8 k Da),并优化了LMWH制备的工艺参数。通过对p H、降解温度、降解时间和H2O2浓度四个因素的单因素试验和响应面分析,结果表明:当p H7.1、温度60℃、降解时间4.5 h、H2O2浓度6.4%时,LMWH的平均分子量为4 050 Da,2~8 k Da的含量为88.57%,抗-FXa效价为103.42 IU·mg^-1,抗-FXa/抗-FIIa值为3.34,符合市场对LMWH的要求。为磁化水结合H2O2制备LMWH提供了良好的试验基础。
The fine heparin as raw material was used to prepare low molecular weight heparin (LMWH,2-8 kDa) by hydrogen peroxide with magnetized water, and the process parameters for preparation of LMWH was optimized. Four factors of pH, degradation temperature, degradation time, concentration of H2O2 were tested by single factor and response surface experiments analysis. The results showed that the optimum condition of LMWH preparation were pH 7.1, degradation temperature 60 ℃, degradation time 4.5 h, and concentration of H2O2 6.4%. Under such conditions, the average molecular weight of LMWH was 4 050 Da, content of 2-8 kDa was 88.57%, titer of anti-FXa was 103.42 IU·mg^-1, anti-Fxa/anti-FIIa was 3.34. These indicators of LMWH met the requirements of mart, which provide an useful experimental foundation for preparation of LMWH by hydrogen peroxide with magnetized water.
