摘要
目的探讨膜联蛋白A1(Annexin A1、ANXA1)和P19蛋白与胆囊癌发生、发展的关系。方法应用免疫组织化学和Western blot法检测术前未经化疗的56例胆囊癌术后标本(癌组织和癌旁组织)及56例非肿瘤性胆囊组织标本中的Annexin A1和P19蛋白。结果免疫组化结果提示:ANXA1在胆囊癌组织、癌旁组织及非肿瘤性胆囊组织中阳性表达率分别为12.50%(7/56)、26.78%(15/56)、69.64%(39/56),差异有统计学意义(P<0.01)。P19在三种组织中阳性表达率分别83.92%(47/56)、62.50%(35/56)、25.00%(14/56),三者间差异有统计学意义(P<0.01)。Western blot结果进一步证实:胆囊癌中ANXA1蛋白表达量高于癌旁组织及非肿瘤胆囊组织(P<0.05),P19蛋白在胆囊癌中的表达低于癌旁组织及非肿瘤胆囊组织(P<0.05)。ANXA1和P19在胆囊癌组织中表达与胆囊癌的病理分化类型、临床分期和淋巴结转移有关(P<0.01或<0.05)。胆囊癌中ANXA1和P19蛋白表达呈负相关(r=-0.336,P<0.05)。结论 ANXA1、P19与胆囊癌的发生、发展和淋巴结转移有关,有望成为判断胆囊癌临床病理进程的辅助指标。
Objective To detect the expression levels of Annexin A1 and P19 protein in non- carcinoma gallbladder tissues,gallbladder carcinoma and its surrounding tissues and to explore their relationship with occurrence and development of gallbladder carcinoma. Methods The expression levels of ANXA1 and P19 protein in 56 patients with gallbladder Carcinoma who did not undergo chemotherapy before surgery and 56 patients with non- carcinoma gallbladder tissue were detected by immunohistochemical SP method and Western blotting method. Statistical analysis was performed with reference to the related clinical and pathological features.Results The positive rates of Annexin A1 expression in gallbladder carcinoma,tissue near by non- carcinoma and carcinoma gallbladder tissue were 12. 50%( 7 /56),26. 78%( 15 /56) and 69. 64%( 39 /56),respectively. There was significant difference in the positive rates among the groups. The positive rates of P19 expression in the three tissues were 83. 92%( 47 /56),62. 50%( 35 /56),25. 00%( 14 /56),respectively,and the positive expressions of P19 among the three groups had statistically significant difference. The positive expression of ANXA1 and P19 were correlated with the pathological grades,clinical stages and lymph node metastasis of gallbladder carcinoma. There was negatively correlation between the expression rate of ANXA1 and that of P19 in gallbladder carcinoma. Conclusion The positive expression of ANXA1 and that of P19 may be correlated with the carcinogenesis,development and lymph node metastasis of gallbladder carcinoma.They may serve as an auxiliary marker of gallbladder carcinoma. The combined use of the above markers possibly has certain reference significance in evaluating the malignancy of gallbladder carcinoma and its prognosis. ANXA1 may become a molecular marker of early cancer action of gallbladder carcinoma.
