摘要
目的:建立高效液相色谱法同时测定高油脂火锅底料中的苯甲酸和山梨酸的方法。方法:样品经热碱溶解,超声提取,蛋白沉淀,中性调节,离心分层,滤膜过滤,高效液相色谱测定得到高油脂火锅底料中苯甲酸和山梨酸含量,并对样品预处理条件、色谱条件进行优化选择。结果:苯甲酸含量在1~50μg/ml、山梨酸含量在1~50μg/ml范围内线性关系良好(苯甲酸、山梨酸的相关系数分别为0.9997、0.9997),苯甲酸、山梨酸方法的相对标准偏差分别为4.8%、4.3%,检出限分别为0.72 mg/kg、0.78 mg/kg,回收率分别为87.2%~100.7%、91.6%~96.9%。结论:方法具有操作方便快速、样品净化完全、干扰物质去除干净等特点,灵敏度高,分离度好,回收率高,适合快速测定高油脂火锅底料中的苯甲酸和山梨酸的含量。
Objective: Simultaneous determination of high oil Hot pot bottom material of benzoic acid and sorbic acid to establish a method of HPLC. Methods: The sample after alkali dissolution,ultrasonic extraction,protein precipitation,neutral adjustment,centrifugal separation,membrane filtration,high performance liquid chromatography determination of the steps to get the benzoic acid and sorbic acid in high oil bottom material of chafing dish,and the sample pretreatment conditions and chromatographic conditions were optimized.Results: The method of benzoic acid content in 1 ~ 50 μ g/ml,sorbic acid content in the 1 ~ 50 μg/ml linear relation in the range of good( correlation coefficient of benzoic acid,sorbic acid were respectively 0. 9997 and 0. 9997),the relative standard deviation of benzoic acid,sorbic acid method was 4. 8% and 4. 3% respectively,the detection limit were 0. 72 mg/kg,0. 78 mg/kg,recovery rate were87. 2% ~ 100. 7%,91. 6% ~ 96. 9%. Conclusion: The method has the advantages of convenient operation and rapid,sample clean- up completely,interfering substances are removed,the method is of high sensitivity,good degree of separation,the recovery rate is high,suitable for fast determination of high oil Hot pot bottom material of benzoic acid and sorbic acid content.
