摘要
作者应用杂交瘤技术获得了四株抗黄疸出血群赖型017株钩体外膜的单克隆抗体。经免疫扩散法鉴定表明,其抗体1A7E7为IgG_2,2F9D4、1A7H12和2G3H5为IgG_1。用免疫印迹分析发现,1A7E7特异地识别017株钩体外膜42kd蛋白区带,2F9D4识别三株致病性钩体外膜42kd的蛋白区带,而2G3H5和1A7H12特异地识别三株致病性钩体外膜的31kd蛋白区带。进一步研究致病性钩体所具有的蛋白质抗原分子的性质,对于寻找特异性诊断和保护性抗原会提供一定的线索。
Four McAbs (1A7E7, 2F9D4, 2G3H5,1A7H12) against outer membrane antigensof L. interrogans serovar Lai strain 017 wereproduced by hybridoma technique. McAb1A7E7 was identified to be IgG_(2a), the restIgG_1 by immunodiffusion. Outer membraneantigens of three pathogenic leptospiralstrains (017, 601, 156) and two non-pathogen-ic leptospiral strains (Patoc Ⅰ, 3055) wereanalysed by SDS- polyacrylamide gel elec-trophoresis (SDS-PAGE) and immunoblot-ting with McAbs.It was found that McAb1A7E7 recognized specifically the 42kd an-tigenic band of strain 017; McAb 2F9D4 the42kd antigenic bands of strain 017, 601,156, and McAbs 2G3H5 and 1A7H12 the 31kd antigenic bands of 017, 601,156. Fur-ther study of antigenic properties possessedby pathogenic leptospira may provide somenew clues to look for specific diagnostic andprotective antigens.
