用HPLC法同时测定扶正平消胶囊中龙胆苦苷和迷迭香酸的含量

Simultaneous determination of gentiopicroside and rosmarinic acid in Fuzhengpingxiao capsules by HPLC method

目的：建立HPLC法同时测定扶正平消胶囊中龙胆苦苷和迷迭香酸的含量。方法：采用Agilent Eclipse XDB-C18色谱柱（250mm×4.6mm,5μm）,以0.1%甲酸（A）-乙腈（B）为流动相进行梯度洗脱,0-5min 88%A,5-10min88%-82%A,10-15min 82%-70%A,15-20min 70%-60%A;流速1.0ml/min;柱温25℃;检测波长280nm。结果：龙胆苦苷在6.75-431.6μg/ml（r=1.000）,迷迭香酸在3.16-201.8μg/ml范围内呈良好线性（r=1.000）。日内和日间精密度均〈2%（n=3）。平均加样回收率分别为（100.89±2.25）%和（98.19±1.13）%（n=6）。结论：该方法简便、结果准确、重复性好,可用于扶正平消胶囊中龙胆苦苷和迷迭香酸的质量控制。

Objective：To establish a HPLC method for simultaneous determination of gentiopicroside and rosmarinic acid in Fuzhengpingxiao capsules.Methods：Agilent Eclipse XDB-C18column（250mm×4.6mm,5μm）was used for separation.Mobile phase was composed of 0.1%formic acid（A）-acetonitrile（B）in gradient elution at a flow rate of 1.0ml/min：0-5min88% A,5-10 min 88%-82% A,10-15 min 82%-70% A,15-20 min 70%-60% A.The column temperature was 25 ℃ and the detection wavelength was 280 nm.Results：The calibration curves of gentiopicroside and rosmarinic acid showed good linearity in the ranges of 6.75-431.6μg/ml（r=1.000）and 3.16-201.8μg/ml（r=1.000）,respectively.The results of intra-day and interday precisions were both within 2%（n=3）.The average additional recovery rates were（100.89±2.25）% and（98.19±1.13）%（n=6）,respectively.Conclusion：The HPLC method proves to be simple,accurate and reproducible.It could be used for quality control of gentiopicroside and rosmarinic acid in the preparation of Fuzhengpingxiao capsules.
