摘要
目的:探讨黄芩素(Baicalein)对人乳腺癌MDA-MB-231细胞肺转移的影响及相关的作用机制。方法:建立人乳腺癌MDA-MB-231细胞裸鼠肺转移模型并给与黄芩素药物干预。实验分为四组:1.对照组(给予生理盐水);2.实验组(给予黄芩素100mg·kg-1);3.低剂量预防组(黄芩素50mg·kg-1);4.高剂量预防组(黄芩素100mg·kg-1)。采用免疫组化SP法和Western blot法,检测裸鼠的肺组织中SATB1,Wnt1,β-catenin,E-cadherin,Vimentin和Snail蛋白的表达。结果:免疫组化结果表明黄芩素可以抑制SATB1,Wnt1,β-catenin,Vimentin,Snail蛋白的表达和促进E-cadherin蛋白的表达。Western blot结果表明黄芩素下调SATB1,Wnt1,β-catenin,Vimentin,Snail蛋白的表达和上调E-cadherin蛋白的表达。各组蛋白平均表达水平差异均有统计学意义(P<0.05)。结论:黄芩素(Baicalein)抑制乳腺癌MDA-MB-231细胞的肺转移,可能机制是抑制SATB1蛋白的表达,阻止Wnt1/β-catenin信号通路,抑制EMT的发生,从而减少乳腺癌肺转移的发生。
Objective: To investigate the inhibition mechanism of baicalein on the lung metastasis of breast cancer cell MDA-MB-231 in vivo. Methods: An in vivo model of nude mouse of breast cancer with lung metastasis using breast cancer cell MDA-MB-231 were established. Baicalein intervention was conducted. The mice were divided into four groups:1. control group( dealt with normal saline); 2. experiment group( Baicalein 100 mg·kg- 1); 3. low dose preventive group( Baicalein 50 mg·kg- 1); 4. high dose preventive group( Baicalein 100 mg·kg- 1). Immunohistochemistry and Western blotting analysis were adopted to detect the expression of SATB1( special AT rich sequence binding protein1),Wnt1,β-catenin,E-cadherin,Vimentin and Snail. Results: Immunohistochemistry results showed that baicalein could inhibit the expression levels of SATB1,Wnt1,β-catenin,Vimentin and Snail,while promote the expression in of E-cadherin. Western blotting analysis showed that baicalein could decrease the expression level of SATB1,Wnt1,β-catenin,Vimentin and Snail,while increase the expression level of E-cadherin with statistically significant difference. Conclusion: Baicalein may inhibit lung metastasis of breast cancer cell MDA-MB-231 by decreasing the expression of SATB1 and regulating the Wnt1 /β-catenin signaling pathway,and then preventing the occurrence of EMT,suggesting that baicalein may be a potential therapeutic agent for lung metastasis of breast cancer.
