摘要
目的建立丙型肝炎病毒八种亚型的液相芯片分型检测方法。方法通过建立多重PCR扩增、核酸探针偶联及杂交检测方法,建立液相芯片分型检测方法,并评价所建立的方法。结果建立的基于液相芯片技术的丙型肝炎病毒分型检测方法能对八种亚型进行检测和分型,具有较高的特异性和敏感性。对142份临床样本的检测结果表明,该方法具有高通量、快速、敏感、特异的特点。结论本研究建立的方法能实现对丙型肝炎病毒八种亚型的同时快速检测,是丙型肝炎病毒分型检测的一种新方法。
Objective This article establishes a liquichip method which can be used for genotyping eight subtypes of HCV simultaneously. Methods A liquichip method for simultaneously genotyping of eight subtype of HCV is developed,including multiplex PCR method,nucleic probe coupling with microspheres,and hybridization methods. The sensitivity,specificity and accuracy of the method are evaluated. Results The method based on liquichip can genotyping eight subtype of HCV simultaneously. One hundred and forty two serum samples testing results show that the genotyping method has a high specificity,sensitivity and accuracy. Conclusion This method can be used for genotyping eight subtype of HCV simultaneously and quickly,providing a new method for the genotyping of HCV.
