母鼠孕中期暴露壬基酚对子代大鼠海马组织形态的影响 被引量:1

Effects of nonylphenol on theneuron structure of hippocampus in offspring rats in gestation

摘要 目的探讨母鼠孕中期暴露壬基酚(NP)对子代大鼠海马组织结构的影响。方法性成熟大鼠按雌雄2∶1合笼饲养交配,再将交配成功的28只母鼠随机分成4组:对照组和低、中、高剂量组,从孕期第9天到第15天空腹灌胃NP,剂量分别为0、50、100、200mg/(kg·d)。剖杀60d龄仔鼠,取海马组织制片,HE染色、尼氏染色及在电子显微镜下观察海马区神经元组织结构的改变,图像处理系统测定海马区尼氏体积分吸光度值(IA)。结果与对照组比较,低剂量组皮质区神经元出现固缩;中、高剂量组海马尼氏体的IA均明显降低(P<0.05),高剂量海马区神经元固缩为三角形或多角形,核深染;中、高剂量组神经元细胞均匀化改变,内质网多,细胞普遍水肿,周围组织水肿。结论母鼠孕中期暴露100mg/(kg·d)的NP可致仔鼠海马组织神经元的组织形态构发生改变。 Objective The purpose of this study was to explore whether mid-gestation exposure to nonylphenol(NP)would induce the alterations in the neuron structure of hippocampus in offspring rats.Methods Each male rat was mated with 2female rats in one cage.Twenty eight dams were divided into four groups randomly,which were administrated with NP at doses of 0,50,100,200mg/kg/day daily in gestational day from 9to 15.Hippocampus tissues in pups were taken to observe the alterations in the neuron structure of hippocampus on PND 60.Integrated optical density(IOD)was measured by image processing system.Results Compare with the control group,there was pyknosis in the neuron of cortical area in the low treatment group;IOD of nissl body decreased significantly in both middle and high groups(P〈0.05).The neuron in hippocampus was karyopynosised present on triangles or polygons,stained darkly.Neuronal cells in the middle and high group were changed uniformly.Edema was present in cells and surrounding tissue.Conclusions Dams gestationally exposed to NP in the dose of 100mg/kg/day might alter the neuron structure of hippocampus in offspring rats.
