芪白平肺胶囊对COPD痰瘀阻肺证肺血管重构大鼠Rho激酶表达的影响

Influence of Qibai Pingfei Capsule on expression of Rho-kinase in pulmonary vascular remodeling of COPD rats with syndrome of phlegm and blood stasis obstructing lung

目的:观察芪白平肺胶囊对COPD痰瘀阻肺证大鼠肺血管重构大鼠Rho表达的影响,并探讨其作用机制。方法:健康SD雄性大鼠100只,随机分组为正常组、模型组、芪白平肺低剂量组、芪白平肺高剂量组、法舒地尔组,采用参照文献造模方法,建立慢性阻塞性肺疾病痰瘀阻肺证病证结合的COPD肺血管重构大鼠模型。药物干预后使用ELISA测定各组大鼠血清中Rock-1的含量,采用Western Blot技术比较各组大鼠Rock-1以及p-MYPT-1蛋白表达以及RT-PCR测定Rock-1 m RNA的转录水平。结果:模型组血清Rock-1,肺组织中Rock-1 m RNA的转录水平,Rock-1以及p-MYPT-1蛋白表达均显著增加(P<0.01);各用药组大鼠Rock-1以及p-MYPT-1表达水平均有不同程度的下降(P<0.01,P<0.05),以高剂量组为优。结论:Rock-1、Rock-2参与肺血管重构过程。芪白平肺胶囊能有效减轻肺血管重构,其机制可能与其调节Rho激酶在血清、肺组织及肺动脉中的表达相关。

Objective：To observe the influence of Qibai Pingfei Capsule on expression of Rho-kinase in pulmonary vascular remodeling of COPD rats with syndrome of phlegm and blood stasis obstructing lung,and explore the action mechanism.Methods：One hundred male SD rats were randomly divided into control group,model group,low and high doses of Qibai Pingfei Capsule groups and fasudil group,and with 20 rats in each group.The pulmonary vascular remodeling of COPD rat models with syndrome of phlegm and blood stasis obstructing lung were established according to XIAO Shi-liang and LI Ze-geng＇s model establishing method.After intervening with medicines,the contents of Rock-1 in serum of rats were determined by using ELISA,and the protein expression of Rock-1 and phosphorylated MYPT-1 was tested by using Western Blot method.The mRNA expression of Rock-1 was determined by using RT-PCR.Results：The contents of Rock-1 in serum,expression of Rock-1 mRNA expression of Rock-1 and phosphorylated MYPT-1 in lung tissue of rats in model group were increased significantly（P〈0.01）.The expression of Rock-1 and phosphorylated MYPT-1 in lung tissue of rats in treatment groups was decreased to some extent（P〈0.01,P〈0.05）,and the high does of Qibai Pingfei had the best effect.Conclusion：Rock-1 and Rock-2 involved in the process of pulmonary vascular remodeling,and Qibai Pingfei Capsule could relieve pulmonary vascular remodeling effectively.The mechanism might relate its effects on regulating the expression of Rho-kinase in serum,lung tissue and pulmonary artery.
