芍药甘草汤对慢传输型便秘大鼠ICC及肠神经递质的影响 被引量:40

Effects of Shaoyao Gancao Decoction on interstitial cells of Cajal and enteric neurotransmitter in slow transit constipation rats

摘要 目的:观察芍药甘草汤对慢传输型便秘大鼠Cajal间质细胞(ICC)及肠神经递质的影响。方法:50只Wistar大鼠按照随机数字表法分为正常组、模型组、莫沙必利组、芍药甘草汤低剂量组和芍药甘草汤高剂量组,除正常组外,均用复方地芬诺酯灌胃建立慢传输型便秘大鼠模型,给药后检测大鼠粪便数量、含水率,ICC及NO、NOS、VIP、SP等肠神经递质的变化。结果:芍药甘草汤可以增加慢传输型便秘大鼠粪便数量,提高粪便含水率(P<0.05);减少抑制性神经递质NO、NOS和VIP水平(P<0.05),对SP含量有一定程度提高,但差异无统计学意义;提高c-kit蛋白的表达(P<0.05)。结论:芍药甘草汤可以改善便秘症状,其机制可能与降低抑制性神经递质NO、NOS、VIP,增加ICC的数量和功能,进而提高肠道平滑肌的运动有关。 Objective:To observe the effects of Shaoyao Gancao Decoction on interstitial cells of Cajal(ICC) and enteric neurotransmitter in slow transit constipation rats.Methods:Fifty Wistar rats were randomly divided into five groups:control group,model group,mosapride group,Shaoyao Gancao Decoction low-dose group and Shaoyao Gancao Decoction high-dose group.The slow transit constipation rat model was established with compound diphenoxylate by gavage.After treatment,the changes of the volume and moisture content of feces were obversed and the contents of enteric neurotransmitters,such as NO,NOS,VIP and SP,were detected.Results:Shaoyao Gancao Decoction could increase the volume of feces as well as moisture content of feces in slow transit constipation rats(P〈0.05);Shaoyao Gancao Decoction could decrease the contents of inhibitory neurotransmitter,such as NO,NOS and VIP(P〈0.05);Shaoyao Gancao Decoction could increase slightly the contents of SP,so there was no statistical difference between the treatment group and the control group;Shaoyao Gancao Decoction could increase the expression of c-kit protein(P〈0.05).Conclusion:Shaoyao Gancao Decoction could improve symptoms of constipation,which might be related to down-regulate the inhibitory neurotransmitter,such as NO,NOS and VIP,and up-regulate the number and function of ICC that would contribute to the movement of intestine.
