棘霉素对KBV200细胞作用的初步研究

摘要 目的初步研究棘霉素(echinomycin)对KBV200细胞(耐长春新碱的口腔癌细胞)的作用及其作用机制。方法采用MTT法及流式细胞仪分别检测KBV200细胞的增殖和凋亡;用同位素掺入、Western blot、RT-PCR等方法分别检测鞘氨醇激酶、P-糖蛋白、缺氧诱导因子的表达。结果棘霉素可诱导KBV200细胞的凋亡,流式结果出现DNA亚二倍体凋亡峰;棘霉素对鞘氨醇激酶、P-糖蛋白、缺氧诱导因子的表达有不同程度的抑制作用。结论棘霉素对KBV200细胞有细胞毒作用,其作用机制与抑制鞘氨醇激酶、P-糖蛋白、缺氧诱导因子的表达有关。 Objective To research the action and mechanism of echinomycin in KBV200 cell line.Methods The proliferation and cell cycle of KBV200 cells were determined by MTT assay and flow cytometry,respectively.TLC,Western-blot and RT-PCR were used to detect the expressions SPK,P-gp and HIF-1α on KBV200 cells,respectively.Results Echinoderm can induce apoptosis of KBV200 cells,and have different degrees of inhibition effect on expression of SPK,P-gp,HIF-1.Conclusion Echinomycin has better the cytotoxic effect on KBV200 cells,and the mechanism of action is related to inhibition of expression SPK,P-gp and HIF-1α.
