摘要 采用碱性蛋白酶酶解缢蛏,以多肽含量为指标,在酶解p H值、料液比、温度、酶添加量、时间的单因素试验基础上,通过正交试验优化缢蛏蛋白酶解工艺。结果表明,利用碱性蛋白酶对蛋白质进行水解,多肽含量高。碱性蛋白酶酶解缢蛏制备多肽最优工艺参数为:p H值9.5、酶添加量1.5%、料液比为1∶3.0、温度45℃、时间2.5 h,在此最优工艺参数条件下每克鲜缢蛏可提取多肽(81.3±0.4)mg。 This study aims at optimizing the hydrolyzation conditions of peptides extraction from Sinonovacula con^tricta by alkaline protease. The technological parameters such as enzymolysis pH value, the solld-llquld ratio, the hydrolysis temperature, the dosage of alkaline proteasc and the hydrolysis time were subjected to the single factor tests. The orthogonal experiment was conducted. The yield of peptides was investigated to evaluate the effect of enzyme hydrolysis. The results showed that hydrolyzation of protein by al- kaline protease increased the yield of polypeptidc. The optimal hydrolytic conditions were as follows : pH was 9.5, the dosage of al- kaline protease was 1.5%, the solid-liquid ratio was I : 3.0, the hydrolysis temperature was 45 cC and the hydrolysis time was 2.5 hours. Under the optimal conditions of this processing, the yield of peptidee was (81.3+0.4) mg· g-1.
