决银方对HaCaT细胞增殖及细胞周期的影响

Effects of Jueyinfang on the Cell Proliferation and Cell Cycle of HaCaT Cells

目的观察决银方对HaCaT细胞增殖及细胞周期的影响。方法用CCK-8法检测1%,2%,5%和10%浓度决银方作用HaCaT细胞后细胞增殖的变化,以相同体积的DMEM作为空白对照;用流式细胞仪检测1%,2%,5%和10%浓度决银方作用HaCaT细胞后细胞周期的变化,以相同体积的DMEM作为空白对照。结果 CCK-8法检测显示5%浓度的决银方作用HaCaT细胞24h,细胞增殖受到明显抑制(P<0.05);流式细胞仪检测显示,5%浓度的决银方作用于HaCaT细胞24h,G_1期细胞比例明显减少(P<0.05)。结论 5%浓度的决银方对HaCaT细胞增殖有明显抑制作用,其主要作用在细胞周期的G_1期。

Objective To investigate the influence of Jueyinfang on the cell proliferation and cell cycle of HaCaT cells. Methods HaCaT cells were cultured with 1% ,2% ,5%, 10% concentrations of Jueyinfang. CKK-8 assay and flow cytometry were performed to detect the proliferation and cell cycle of HaCaT cells respectively, using DMEM of the same volume as blank control. Results CKK8 assay showed that proliferation of HaCaT cells was obviously suppressed after being cultured with Jueyinfang of 5% for 24 hours （P 〈 0. 05）. Flow cytometry asssy showed the percentage of HaCaT cells at G1 phase decreased obviously （ P 〈 0. 05 ） after 24 hours treatment with Jueyinfang of 5%. Conclusion The 5% Jueyinfang showed an obvious inhibitive effect on the proliferation of HaCaT cells, mainly affecting G1 phase of HaCaT cells.
