摘要
目的测定MiR-155在半抗原(2,4-二硝基氟苯)诱导C57BL/6小鼠特应性皮炎模型皮损中的表达及血清IL-4相关性。方法小鼠造模第1天背部使用0.5%DNFB外涂,第5,8,11,14天耳部及背部外涂0.2%DNFB,14天后48h内处死,后检测皮肤MiR-155相对表达量及血清IL-4的表达,并做Pearson相关性分析。结果模型组皮肤表现出明显炎症,耳部明显肿胀,病理提示表皮明显增厚、真皮炎症细胞浸润,血清检测IgE,IL-4含量明显升高(P<0.05),皮损内MiR-155相对表达量明显升高(P<0.01),皮损MiR-155相对表达量与血清IL-4呈正相关(R=0.99,P<0.01)。结论 MiR-155可能参与特应性皮炎的发病,并可能在以分泌IL-4为主的Th2细胞介导的免疫反应中发挥作用。
Objective To detect MiR-155 in lesions of atopic dermatitis induced by hapten of DNFB on a background of C57BL/6 and explore the correlation of serumal IL-4 level. Methods Topically apply the 0. 5% DNFB (day 1 ) on the dorsal skins and 0. 2% DNFB (5, 8, 11, 14 days) on the ears and dorsal skins, to test the MiR-155 expression and correlation with IL-4 in serum. Results The inflammation of the model group was obvious, and the histopathology of skin lesions showed thickness of the epidermis and the dominant infiltration of lymocytes in the dermis, and serum IgE and IL-4 were elevated significantly. The expressions of MiR- 155 were elevated in the lesions and were positively correlated with IL-4 in serum. Conclusion MiR-155 may be involved in the onset of atopic dermatitis, and may play a role in the Th2 mediated immune response featured by increased IL-4 secretion.
