新疆塔里木马鹿茸c-myc基因cDNA克隆和bHLHZIP结构分析及表达 被引量:2

摘要 采集6头成年马鹿生长60 d茸组织,获得塔里木马鹿茸c-myc基因cDNA全长序列,并比对分析其转录因子的序列特征;结果表明:得到塔里木马鹿茸c-myc基因cDNA 1 840 bp序列,其中ORF区长1 314 bp,编码438个氨基酸;马鹿茸c-myc序列比对,与牛和羊序列相似性最高分别为96%和97%;氨基酸比对分析,在序列121位处马鹿比牛、羊、猪、人类及鼠少1个天冬氨酸(D);马鹿茸c-myc基因的DNA结合域由bHLHZIP结构组成,共80个氨基酸残基,在bHLH区的第16和46位氨基酸处,塔里木马鹿分别为天冬氨酸D和精氨酸R,而牛、羊、猪、人和鼠为天冬酰胺N和赖氨酸L。6个DNA结合位点和E-box的结合位点没有氨基酸变异;羊、牛、猪、人、鼠和马鹿的cmyc基因的Zip序列变异较大。本实验通过0.8 mmol/L IPTG诱导,2 h后构建的Pet-28b-c-myc原核表达载体开始表达目的基因,PAGE检测获得约48 ku目的蛋白质,同时发现随着诱导时间延长没有发现表达量的变化。
