体外成球实验筛选三阴乳腺癌干性相关microRNA实验研究 被引量:5

Spheroid Study in Vitro on Screening the microRNA Associated with Breast Cancer Stem Cells of Triple Negative Breast Cancer

摘要 目的筛选三阴乳腺癌(triple negative breast cancer,TNBC)干性相关microRNA(miRNA),为TNBC不良预后预测提供干性靶标。方法贴壁培养人乳腺癌高转移潜能细胞系MDA-MB-231进行体外成球实验,分别抽提Spheroid和Attachment培养细胞总RNA,利用miRNA芯片比较两种培养细胞表达miRNA的差异情况。结果 Spheroid及Attachment细胞表达差异的miRNA共有157个,其中10个miRNA表达差异最明显,且差异倍数达4倍以上。结论乳腺癌干细胞中差异表达的miRNA可能与TNBC耐药相关,可作为诊断乳腺癌和评判预后的重要靶标,并为抵抗乳腺癌耐药提供可能靶点。 Objective To screen and identify the miRNA associated with breast cancer stem cells by the miRNA ar- ray technique, and predict miRNA associated with drug resistance of TNBC, in order to provide target gene for poor prognosis prediction of TNBC. Methods Total RNA was respectively extracted from Spheroid and attachment culture MDA-MB-231, and the experiment was conducted in vitro. Results Spheroid and attachment culture cell expressed a total of 157 miRNA with differences, including 10 miRNAs with the most significant differences at 4 times or over. Conclusion Differentially expressed miRNAs in stem cells of breast cancer may be related to drug resistance of TNBC. It can be used as an important cri- terion in diagnosis of breast cancer and provide a target point for drug resistance of TNBC.
